OECD 311 Anaerobic Biodegradability of Organic Compounds in Digested Sludge

For sewage sludge digestion conditions, OECD 311 test guidelines are used for the assessment of anaerobic biodegradation of organic chemicals. OECD 311 test guidelines (Anaerobic biodegradation-testing) examine if a chemical can be degraded anaerobically or not, while the OECD 301 series for aerobic biodegradation tests answers whether or not a chemical is degraded by air. The strongest evidence of anaerobic degradation is the presence of biogas.     Introduction of OECD 311

    Organic content in sewage sludge digestion circumstances, anaerobic degradation is measured by the OECD 311 test guideline. OECD 311 test and unlike the aerobic biodegradation tests (OECD 301 series), the other will not determine if something can degrade in oxygen-free environments, e.g., anaerobic digesters, sediments, and groundwater. The global performance characteristic of anaerobic biodegradation is biogas production, mainly methane (CH₄) and carbon dioxide(CO₂). It is a critical aspect of the understanding of chemicals in wastewater treatment plants and natural anaerobic environments as a whole if chemicals degrade anaerobically.

    Test Method of OECD 311

    Test procedure In this test, the amount of carbon (inorganic and methane) that developed from the biodegradation of the test substance is determined by measuring net gas production and net inorganic carbon development in the liquid phase (considering the blank control values). The percentage of biodegradation is determined as the amount of total inorganic carbon and methane-C developed with the measured or estimated amount of carbon from the test substance.

    OECD 311 Sample Preparation

    Specimen DetailsThe washed digested sludge, containing low concentrations of inorganic carbon (<10 mg/L), is diluted to achieve a total solids concentration between 1 g/L and 3 g/L. The test substance is added to the prepared sludge at concentrations ranging from 20 to 100 mg of carbon per liter. 
    Incubation conditionThe mixture is then incubated in sealed vessels at a constant temperature of 35 ± 2°C for up to 60 days.

    OECD 311 Results and Interpretation

    After this testing, biogas production is measured as the increase in headspace pressure in the sealed containers. The inorganic carbon concentration in the liquid phase is also measured at the end of the experiment. Biodegradation is calculated by taking the total amount of carbon produced (in methane and inorganic carbon) and expressing it as a percentage of the initial carbon content of the test material. A high percentage of conversion of carbon into biogas indicates that the test material is readily biodegradable under anaerobic conditions. A lower percentage indicates that the anaerobic biodegradability may be limited, and further environmental risk assessments may be needed.
