OECD 120 Introduction

OECD 120 refers to Test No. 120 under the OECD Guidelines for the Testing of Chemicals, which is titled “Solution Extinction Behaviour (Solubility in Water).”  The goal of OECD 120 is to provide a guideline for the determination of solubility of a chemical substance in water. Solubility in water is a vital property that affects the environmental fate, toxicity, and the potential aquatic bioaccumulation of chemicals.

Test procedure for one sample sizeFor a direct analysis of the aqueous phase, one sample size is sufficient. Analysis of the aqueous phase for the total polymeric species is possible either by a sufficiently sensitive method or by vacuum evaporation to dryness of the aqueous extract and spectroscopic or AAS/ICP analysis of the residue.
Test procedure for two different sample sizes.  All aqueous extracts are analysed for total organic carbon (TOC).A gravimetric determination is performed on the undissolved/not-extracted part of the sample.

OECD 120 Equipment and Sample Preparation

Specimen Details3 samples of 10g particles test substance are weighed into each of three vessels fitted with glass stoppers. 1000 ml of water is added to each vessel.
Specimen PreparationThe vessels are tightly stopped and then agitated at 20 °C, normally for 24 hours. Then, the content of each vessel is centrifuged or filtered, and the concentration of polymer in the clear aqueous phase is determined for biodegradability. 

OECD 120 Results and Interpretation

The solubility in mg/L or ug/L at a specific temperature is determined.  The results show the extent of polymer dissolution or extraction in the water. A larger amount of material in the aqueous phase suggests the polymer dissolves or extracts to a greater extent. The relative standard deviation (RSD) will tell you how reproducible the results are.
