Preparations for Microscope Calibration

In order to block any interference, the entire microscope(lenses,stage,and eyepieces) should be thoroughly clean. To optimize the precision of your calibrations measurements, let the microscope stay for at least 30 minutes in room temperature. Make sure the items in the stage, focus knobs, illumination system, and other components of the microscope are working correctly.

Microscope Calibration Process

• To calibrate the microscope’s magnification against individual objectives, the stage micrometer or calibration slide with known dimensions can be used. Using them as a reference make measurement of gaps between calibration markers and compare them to known values.
• In order to acquire more precise focusing at the various magnifications, move the scope focus knobs. In order to gauge the sharpness as well as the clarity of the picture, apply a standard specimen with fine details.
• Adjust illumination levels of the microscope to suit and maintain uniform illumination across the entire view area. To find the light intensity at various levels, use either a calibrated exposure meter or a neutral density filter.

Microscope Calibration Verification and Documentation

• Verification Testing: To ensure that the outcome of the microscope calibration is accurate and analyze the performance of the microscope, there is a need to do other tests, like the resolution testing which targets the resolutions of the instrument.
