ASTM E2180 Introduction
The process also enables the assessment of an antimicrobial treatment’s “shelf-life” or long-term durability. This may be done by evaluating both non-washed and washed samples over a period of time. This is a quantitative test method to check the antimicrobial activity of hydrophobic surfaces. This test standard utilizes an agar slurry inoculumvehicle which facilitates even contact between microorganisms and antimicrobial-treated hydrophobic surfaces
The required incubation period, often 24 hours, is given to the treated and untreated rudders to sit at the proper temperature. To determine the initial concentration of the inoculated slurry, separate controls are immediately neutralised, sonicated, serially diluted, and plated. The infected agar slurry is then adequately removed from the slides using sonication for a certain amount of time, followed by the counting of the surviving organisms. Dilutions are made, then the necessary agar is plated, followed by incubation.

ASTM E2180 Test Method
Test procedure
The test (treated) and control (untreated) samples of hydrophobic substances are covered in an agar slurry that has been infected with test microorganisms. Microorganisms from the control and test samples are collected separately using a neutralizing solution after a contact time of 24 hours. Serial dilutions are created on an agar plate. These plates are then incubated for 48 hours in a setting designed to promote the development of the test organisms. By comparing the decrease of bacterial colonies in treated samples with bacteria colonies found in untreated samples, bacterial colonies are enumerated and antimicrobial activity is estimated.
E2180 Equipment and Sample Preparation
Microbial testing
The tools necessary to carry out microbial testing will include an incubator, containers sterilised, a laminar flow hood, nutrient agar, an inoculum with known bacteria concentration, and a steriliser for a clean environment and for prevention of contamination.
Recovery and Enumeration
Samples are prepared, inoculated with specific organisms, and applied to test pieces. After 24 hours, microorganisms are recovered and plated on agar medium for 48 hours to count bacterial colonies. In this process, both the treated and the untreated samples are subjected to treatment by antimicrobial agents.
ASTM E2180 Results and Interpretation
The test compares treated surfaces with untreated controls for bacterial reduction, quantitative analysis, long-term efficacy, microorganism specificity, and environmental relevance. It also tests the “shelf life” of antimicrobial treatments-evaluation of their effectiveness after different periods in various environmental conditions.
Tests related to ASTM E2180
ASTM E2180 is related to ISO 22196, ASTM E2149, EN 1276, and ASTM G21 are the International standards that have evaluated the antibacterial activity of the materials such as plastics, textiles, and chemical disinfectants based on the reduction in bacterial colon and fungus resistance.
Use of E2180 in Various Industries
ASTM E2180 is one of the laboratory standards used by many industries, from medical devices, consumer products, and textiles to construction materials and food packaging, to test the antimicrobial properties of materials in preventing infections, maintaining efficacy, and guaranteeing product longevity.
Materials Commonly Tested with E2180
The most common tested materials are plastics, vinyl products, medical devices, epoxy resins, textiles, and hard surfaces. The major applications of antimicrobial agents to plastics for microbial reduction are usually treated with various formulations. Vinyl products ensure hygiene for its occupants, medical devices are evaluated for their antimicrobial properties, among others.
Common Challenges and Troubleshooting in ASTM E2180 Testing
The use of an agar slurry inoculum makes it possible for the ASTM E2180 method to overcome issues such as surface tension, preparation of the inoculum, recovery of microorganisms, incubation conditions, and testing biofilm by standardising its preparation, effectively eluting bacteria, and adhering to specific conditions.
Importance of ASTM E2180
This standard is an antimicrobial efficacy quantitation test for flat surfaces, which helps ensure product conformance to hygiene and safety standards as well as regulatory compliance in other industries.
