Test Method of OECD 306

There are two specified methods: the Shake Flask Method and the Closed Bottle Method. The Shake Flask Method allows for a maximum of 60 days, while the Closed Bottle method suggests a maximum of 28 days.

Test procedure for the Snake Flask methodIn this test, the dissolved organic carbon (DOC) removal arising from aerobic biodegradation in a shaking flask mode is measured. The percentage degradation will be calculated as DOC removal divided by DOC dosed into the test medium.
Test procedure for the Closed Bottle methodIn this test, In this assay, the dissolved oxygen (DO) consumption arising from aerobic biodegradation in a closed bottle mode without headspace is measured. The percentage degradation will be calculated as DO consumption divided by the theoretical oxygen demand (ThOD). If the ThOD cannot be determined, then the COD could be experimentally determined and used in a less preferred sense as a substitute.

Specimen Details for the closed bottle method.The test substance is introduced into filled, sealed bottles containing natural seawater, typically at concentrations of 2–10 mg/L. The bottles are incubated in the dark at a constant temperature for 28 days.

Results and Interpretation OECD 306

For the Snake Bottle method, biodegradation is monitored by measuring the decrease in Dissolved Organic Carbon (DOC) over time. A reduction of ≥70% in DOC indicates significant biodegradability. And for the closed bottle method, biodegradation is assessed by measuring the consumption of dissolved oxygen, which reflects microbial activity. A reduction of ≥70% in DOC indicates significant biodegradability.

The snake bottle method is suitable for substances that are highly soluble, non-volatile, and non-adsorbing. The Closed bottle method is suitable for substances that are soluble, volatile, or adsorbing.
