    ASTM E578 test method covers the performance properties of fluorescence measuring systems. The limits of the linearity response are determined with respect to the range of concentration of a fluorescence substance.

    Specimen size should be as per the requirements. This test method can be applied to 10-mm path length cuvette formats and instruments covering a wavelength range within 190 to 900 nm.

    ASTM E578 Testing Procedure and Requirements

    The test apparatus uses a standard test solution of quinine sulfate dehydrate in sulfuric acid to test the linearity of fluorescence measuring systems. Instruments like cuvettes, slits, or filters are used to detect the fluorescence of the test solution in a measuring system.  Multiple readings are taken, and a graph is plotted. The upper and lower limits of linearity are determined and expressed in micrograms per milliliter of the standard solution.

    The difference between the fluorescence intensity and the reading of the blank solution is represented by ‘S.’ ‘F’ represents the observed fluorescence intensity.
