    ISO 9888 Equipment and Sample Preparation

    Specimen DetailsIn this test, BOD bottles are filled with a mineral medium, a concentrated microbial inoculum (e.g., activated sludge or surface water), and a small amount of the test compound to limit toxicity. The bottles are filled up to the brim and capped to prevent an exchange with the atmosphere. Oxygen consumption is followed over 28 days by measuring the Dissolved Oxygen (DO) initially, then at regular time intervals.

    The biodegradation is ultimately determined by calculating the percentage of oxygen utilized concerning the theoretical oxygen demand (ThOD) of the substance. A 60% reduction in oxygen after 28 days suggests that the compound is easily biodegradable in aerobic aquatic environments.

    The low solubility of test substances can restrict microbial access, and toxicity to microorganisms may lead to reduced oxygen uptake; employing a lower concentration or a toxicity control can assist in diagnosing this issue. Variability in inoculum quality can impact the reliability of biodegradation; fresh, active sludge is favored for optimal results. Errors in oxygen diffusion can occur if bottles are not sealed correctly or if there are temperature fluctuations.
