OECD 208 Test for Emergence and Growth of Terrestrial Seedlings

    OECD 208 Introduction

OECD 208 analyzes the effects of chemicals on the germination of seedlings and the early stages of growth of terrestrial plants. It is used to assess the phytotoxicity of a variety of materials, including pesticides, industrial chemicals, and soil contaminants. This test helps identify if a chemical could pose a risk to plants in either agricultural or natural settings.

    OECD 208 Test Method

Test procedure The terrestrial plants are placed in the soil that has been treated with the test substance.

OECD 208 Equipment and Sample Preparation

Specimen DetailsFor the test sample is soluble in water, it is dissolved in water and then mixed with soil. For the test sample is of low solubility, it is dissolved in suitable volatile solvent, such as acetone or ethanol, then mixed with soil. For the insoluble test sample , the chemicals are directly mixed with dry soil.
Specimen PreparationThe most common plants used in this experiment are carrots, sunflowers, lettuce, cucumber, soybean, onion, rice, wheat, corn, and other typical plants. They are planted in natural soils, including sandy loam, loamy sand, sandy clay loam, or in commercially available potting soil equally not exceeding 1.5% organic carbon. Seeds are positioned in contact with soil that has been treated with the test substance and observed for effects typically 14 to 21 days after 50% of the seedlings in the control group have emerged.

    OECD 208 Results and Interpretation

At the end of this test, we assess seedling emergence, biomass measurements, shoot height, and any visible harmful effects on various parts of the plant. Several criteria must be met for the test to be considered valid, including:

    1. A minimum seedling emergence of 70% for the blank control

2.No visible phototoxic effects on the control seedlings

    3. At least 90% mean survival rate of the emerged control seedlings throughout the study     duration
